Coupling Reaction¶
TL;DR¶
Amino acid coupling requires activation of the carboxyl group using carbodiimides (DIC) or uronium salts (HBTU/HATU) with a base. Coupling efficiency directly determines final peptide purity.
Activation Mechanism¶
Fmoc-protected amino acids (Fmoc-AA-OH) must be activated before coupling to the growing peptide chain.

Coupling Conditions¶
Standard Conditions¶
- Solvent: DMF (0.1–0.3 M concentration)
- Temperature: 20–25 °C
- Time: 30–60 minutes
- Mixing: Nitrogen bubbling or gentle shaking
Microwave-Assisted Coupling¶
- Temperature: 50–75 °C
- Time: 5–10 minutes
- Advantage: Reduced cycle time, better for long sequences
- Risk: Racemization at His, Cys residues
Double Coupling¶
For difficult couplings (sterically hindered, β-branched AAs): 1. Perform standard coupling 2. Drain, wash 3. Repeat with fresh reagents

Monitoring Coupling Completion¶
Kaiser Test (Ninhydrin)¶
- Positive (blue/purple): Free amine present → incomplete coupling
- Negative (yellow): Complete coupling
- Sensitivity: ~5 μmol/g free amine
- Note: Does not work for Proline (secondary amine)
Chloranil Test¶
- For Proline and other secondary amines
- Positive (blue): Incomplete
UV Monitoring¶
- Track Fmoc deprotection absorbance at 301 nm
