Rink Amide Resin¶
Solid Support for C-Terminal Amide Peptides¶
Definition¶
Rink amide resin produces peptides with C-terminal amide (-CONH₂), which is the native form for many bioactive peptides.
Specifications¶
| Parameter | Typical Value |
|---|---|
| Base polymer | Crosslinked polystyrene |
| Loading | 0.3–0.8 mmol/g |
| Linker | Rink amide (4-[(2,4-dimethoxyphenyl)(Fmoc-amino)methyl]phenoxy) |
| Swelling (DMF) | 4–6 mL/g |
Key Feature¶
The Rink amide linker incorporates a pre-attached Fmoc-protected amine group. Upon TFA cleavage, the peptide is released with a C-terminal amide.
Cleavage¶
- Reagent: TFA (90–95%)
- Product: Peptide-CONH₂ (C-terminal amide)
- Time: 1.5–3 hours
Why C-Terminal Amide Matters¶
Many naturally occurring peptides (and therapeutic peptides) have C-terminal amidation, which: - Increases resistance to carboxypeptidases - Enhances biological activity - Alters peptide charge and solubility
Common Peptides Requiring Amide¶
- GLP-1 analogs (semaglutide, liraglutide)
- Melanotan II
- Thymosin alpha-1
-
Most therapeutic peptides >10 AA
